Description: Speedy NZYTaq DNA polymerase is an ultrapure recombinant DNA polymerase displaying a faster polymerization reaction than any other conventional non-proofreading enzyme. Speedy NZYTaq DNA polymerase was recently improved for superior performance in fast protocols using a vast range of templates. In addition, conditions to further minimize the running times were also reviewed. Only 5 seconds are required for the successful synthesis of 1 kb size DNA. The enzyme retains its speed (5 sec/kb) when amplifying fragments up to around 2-3 kb. Resulting PCR products have an A-overhang and are suitable for cloning with NZYTech´s NZY-A PCR cloning kit (MB053) or NZY-A Speedy PCR cloning kit (MB137).

Successful amplification in different PCRs with limited extension time (5 sec/kb):
The elongation rate of Speedy NZYTaq DNA polymerase was tested with a range of different-sized DNA fragments  of  different origins using a limited extension period of 5 sec/kb and 2.5 U of enzyme.

Two different batches of Speedy NZYTaq DNA polymerase were used to amplify the following fragments: Lanes 1: 350 bp PCR product (sheep genomic DNA, 80 ng). Lanes 2: 1.3 kb PCR product (E. coli genomic DNA, 50 ng). Lanes 3: 2 kb PCR product (plasmid DNA, 10 ng). Lanes 4: 2.7 kb PCR product (Clostridium thermocellum genomic DNA, 50 ng). Lane M: NZYDNA Ladder III. PCRs were performed in 50 µL reaction mixtures containing 2.5 mM MgCl₂ with elongation times of 1.8 sec, 6.5 sec, 10 sec and 13.5 sec for 350 bp (1), 1.3 kb (2), 2 kb (3) and 2.7 kb (4) fragments, respectively.
