NZYTaq 2× GC-Enhancer Solution

Description: NZYTaq 2× GC-Enhancer Solution was specially developed to overcome difficulties with polymerase chain reaction (PCR) amplification of GC-rich DNA templates using Taq DNA polymerase. The solution contains a proprietary formulation that has proven to be particularly useful when attempting to amplify highly GC-rich DNA sequences (GC% of 60-80%).  In addition, NZYTaq 2× GC-Enhancer Solution can also improve specificity by reducing the amplification of non-specific products generated from GC-repeat sequences. A mixture of NZYTaq 2× GC Enhancer Solution and NZYTaq 5x Optimizer Solution can be used to amplify GC-rich DNA templates when the presence of PCR inhibitors is suspected.

Successful amplification of highly GC-rich templates (>65%):

The addition of 2× GC-Enhancer Solution dramatically improves yields and specificity of problematic targets in PCR amplifications using NZYTaq DNA polymerase. To test the effect of this supplementation, six different human genomic DNA sequences with GC-contents ranging from 65 to 77 % were amplified, in parallel, using NZYTaq DNA polymerase or NZYTaq DNA polymerase plus the enhancer solution.

The six human genomic DNA sequences (1-6) have a GC content of 77.2%, 66.4%, 68.7%, 72.9%, 71.6% and 65.7%, respectively. Results demonstrate a shift from non-specific products and smeared bands on agarose gel to the desired band in a wide range of GC-contents, when 2× GC-Enhancer Solution was used.
