Opis

The plasmid pJL3 encodes chloramphenicol resistance and contains the Bacteriocin Release Protein under control of the lpp lac tandem promoter operator. Moderate induction with IPTG (10 - 40 μM) will give, dependent on the host strain used, a release of periplasmic and certain cytoplasmic proteins. Full induction might give quasi-lysis of host cells. This lysis can be hampered by adding 10 - 20 mM Mg2+ ions to the culture medium (preferably normal broth, i.e. Lab-Lemco broth (Oxoid) supplemented with 0.5% NaCl and 0.5% Na lactate).
